Clone Isolation |
Selection
of high producing cells is currently a major bottleneck in the biomanufacturing
of recombinant proteins and mAbs. The Clonepix FL technology from Genetix allows
for the high throughput screening and selection producers at a much earlier stage
of biomanufacturing production and helps eliminating of much of the manual labor
required by traditional clone selection methods. Screening cell populations for
high producing clones traditionally involves cell sorting, limiting dilution, ring
cloning and even simple manual colony collection. All of these procedures are time
consuming, labor intensive, costly and prone to cross contamination and error.
The ClonePix FL technology uses a target protein specific capture antibodies conjugated
to a fluorophore for detection and selection of producing cells. Build into the
ClonePix instrument imaging software selects high producing clones, typically the
top one to five per cent, and directs automatic robot arm to pick clones off the
microplate for further expansion and analysis.
The
process begins by plating cells in the semi-solid medium. Cells are partially immobilized;
they can grow, multiply and form colonies. Semi solid medium also traps the complex
that forms between the fluorescent capture probe and the
secreted target protein
in the vicinity of colonies. The fluorescently conjugated capture and detection
antibodies can be added to the medium during the time of plating or can be sprayed
over the semi solid medium 24-48 hrs prior to cell picking.
The ClonePix FL system is compatible for both suspension and adherent cells. The
picking process is very gentle; cells retain viability while automatically transferred

Clone isolation
CDI Bioscience clone isolation service is based on ClonePix FL technology - a technology
for screening thousands of mammalian cells and selectively collecting only the highest
value, rare-event clones in a few hours. The broad range of suspension and adherent
cells can be utilized, including hybridoma, CHO, HEK293 and mouse ES cells.
Clone stability screening
CDI Bioscience offers clone stability screening. Selected clones identified with
specific fluorescent marker and level of fluorescents is measured over a period
of time to assure high consistency.
Target gene expression screening
CDI Bioscience can isolate clones based on direct fluorescence from reporter
proteins like GFP. The fluorescence intensity can indicate clones with stable protein
expression or correct protein folding.
Stem cells screening
CDI clone isolation services can be used for stem cell applications. Our technique
allows easily distinguish between differentiated and pluripotent cells. Plates are
imaged with white light, and the relevant clones are detected by florescent markers
of pluripotensy such as anti-SSEA-1.
