| RP Shift® technology |
RP Shift® technology is based on conditionally producing a phenotype change in
biopharmaceutical producing cells in culture. The stable introduction of
full-length cDNA fragments of such genes under control of inducible promoters
will not only stop cell division, but will increase the entire secretory system.
The phenotype change is characterized by an increase in cell volume, a flattened
morphology, increased numbers of mitochondria, expanded endoplasmic reticulum
(increased protein secretion) and increased protein synthesis factors. Such
cells have longer lifespan and are also substantially more resistant to
environmental stresses, such as lowered pH, loss of serum factors, osmotic
changes and other impedance that triggers cell death in proliferating
population. By this virtue RP Shift® increases cell stability and allows higher
concentration of secreted products. CDI has routinely demonstrated 2 to 7 fold enhancement of
the production of protein from fibroblast, Chinese hamster ovary (CHO), mouse myeloma
and mouse hybridoma cells. |
